CC tissues from all rats were collected in liquid nitrogen or embedded in paraffin. Western blot was used to detect the protein level of transforming growth factor β1 (TGF-β1), smad2/3, phospho-smad2/3, collagen I and collagen IV in CC. Immunohistochemical staining and masson’s trichrome staining were conducted to detect the expression of TGF-β1, α-SMA and the ratio of smooth muscle to collagen. In addition, terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was conducted to calculate the apoptosis index (AI) of cells in CC. Besides, the protein content of caspase3, B-cell lymphoma 2 (Bcl-2) and Bcl-2-associated X (Bax) protein were also measured in CC and high concentration glucose treated CC smooth muscle cells (CCSMC) isolated from WTR and TGR. Results: The protein expressions of TGFβ1, smad2, collagen IV and the ratio of phospho-smad2/smad2 were increased in WTDM when compared to the control group, while these parameters were significantly attenuated in TGDM. However, the protective effect of hKLK1 was abolished by HOE140 in TGDMH. Furthermore, the contrary trends of α-SMA content and the smooth muscle/ collagen ratio changes in CC of each group were also observed. No significant difference in collagen I level was found between the five groups. In addition, when compared to the heightened level of AI, caspase3 and Bax/Bcl2 ratio in the CC of WTDM, these parameters were slightly decreased in TGDM. Meanwhile, hKLK1 could effectively inhibit the increase of the apoptosis related indexes in high glucose treated CCSMC in vitro. Conclusions: hKLK1 could exert anti-fibrosis and apoptosis effect in penile tissue of streptozotocin-induced diabetic rats as well as in CCSMC treated with high glucose.

Salvianolic acid B improves insulin secretion from interleukin 1β-treated rat pancreatic islets: The role of PI3K-Akt signaling

Background and Objective: Oxidative stress induced by proinflammatory cytokines such as IL-1β plays a major role in β-cell destruction in diabetes type 1. Salvianolic acid B (Sal B) is a polyphenolic compound with antioxidant and protective effects. Thus, objective of this study was to assess the protection exerted by Sal B on isolated rat islets exposed to IL-1β and to investigate an underlyin...

Insulin resistance in penile arteries from a rat model of metabolic syndrome.

BACKGROUND AND PURPOSE Metabolic and cardiovascular abnormalities accompanying metabolic syndrome, such as obesity, insulin resistance and hypertension, are all associated with endothelial dysfunction and are independent risk factors for erectile dysfunction. The purpose of the present study was to investigate the vascular effects of insulin in penile arteries and whether these effects are impa...

تغییر بیان ژن SHIP2 (SH2 domain containing inositol 5-phosphatase) با استفاده از سیستم رتروویروس در سلول های کبدی HepG2

Introduction: Dyslypydmy is one of the risk factors of cardiovascular disease in diabetics. Dyslypydmy is diagnosed by increasing in plasma triglyceride density, decreasing HDL Cholesterol, and increasing LDL especially small LDL. Several evidences from human and animal studies indicate that the role of insulin resistance is a major cause of hypertrigly ceridemia in diabetics and people with me...

FM19G11 reverses endothelial dysfunction in rat and human arteries through stimulation of the PI3K/Akt/eNOS pathway, independently of mTOR/HIF-1α activation.

BACKGROUND AND PURPOSE FM19G11 up-regulates mammalian target of rapamycin (mTOR)/hypoxia inducible factor-1α (HIF-1α) and PI3K/Akt pathways, which are involved in endothelial function. We evaluated the effects of FM19G11 on defective endothelial vasodilatation in arteries from rats and humans and investigated the mechanisms involved. EXPERIMENTAL APPROACH Effects of chronic in vivo administra...
